Nelly was born in Austin, TX, where his father was serving in the Air Force. He grew up in St. Louis, specifically the suburb of University City. In 1993, he formed a rap group known as the St. Lunatics, consisting of himself, City Spud, Poppa Leezy, and brothers Kyjuan and Murphy Lee. Scoring an independent hit with "Gimme What U Got" in 1997, the St. Lunatics pushed Nelly toward a solo career, believing he had the star power to be signed first and to bring the rest of the group with him. A&R Kevin Law heard his music and signed him to Universal Music Group in 1999, over the objections of colleagues at the label who strongly disliked Nelly's music. The label pushed Nelly's Midwestern roots while promoting him, looking to exploit the novelty of a rapper from St. Louis, propelling his debut album, Country Grammar, to #3 on the Billboard Hot 200. The album spawned several hit singles, and members of the St. Lunatics appeared on the singles "Ride Wit Me" and "Batter Up." The album was eventually certified nine times platinum, and Nelly was a bona fide star.

Following the success of Nelly's solo album, the St. Lunatics immediately went to work on a group album, releasing Free City, their major label debut, on June 5, 2001. The album was certified platinum, and the single "Midwest Swing" cracked the Billboard Hot 100, helping to cement the St. Louis hip hop scene's position in the mainstream.

In 2002, Nelly returned to solo work with his sophomore album Nellyville, which debuted at #1 on Billboard, with singles "Hot In Herre" and "Dilemma" both shooting to #1 on the pop charts and winning Grammys. Riding this wave of enormous success, Nelly released a remix album, Da Derrty Versions: The Reinvention the following year, as well as launching several companies: a sneaker line called the Air Derrty, an energy drink called Pimp Juice, two clothing lines called Vokal (for men) and Apple Bottom (for women). He experienced his first brush with controversy around this time, as Derrty's single, "Tip Drill Remix (E.I.)," received criticism due to misogynistic lyrics. The setback would be short-lived, however, as the singles from the album continued to do well and Nelly's collaboration with Diddy and Murphy Lee for the Bad Boys II soundtrack, "Shake Ya Tailfeather," also hit #1 and won Nelly another Grammy. In 2003 he also founded a bone marrow drive, called Jes Us 4 Jackie, after his sister, Jackie Donahue, was diagnosed with leukemia. The drive hoped to find a bone marrow match that might save Donahue, as well as help to raise money and receive bone marrow donors for others suffering from leukemia.

In 2004, Nelly released two albums, Sweat and Suit, on the same day, with the former featuring a more upbeat hip hop and dance vibe, and the latter being more of an R&B album. The two albums debuted at #2 and #1 respectively on the Billboard 200, and Suit gave Nelly two more top 5 singles in "My Place" (featuring Jaheim) and "Over and Over" (featuring Tim McGraw). During this time period Nelly briefly feuded with fellow St. Louis hip hop star Chingy, although a mutual friend helped end the beef. In 2005, a compilation of Sweat and Suit, aptly titled Sweatsuit, was released with three new tracks, one of which would become another #1 single in "Grillz." He also acted in his first major film role, portraying football-playing convict Earl Meggett in Adam Sandler's remake of The Longest Yard. Tragedy struck this same year, when Donahue lost her battle with leukemia.

After several delays, Nelly finally returned with Brass Knuckles in 2008, which was a mild success compared to his past work. He was preparing to return with another album the following year when his cousin, Michael Johnson, was murdered, delaying the release of the album. Nelly returned in 2010 with 5.0, which featured the top 5 single "Just a Dream," Nelly's biggest hit in five years. His seventh album, M.O., was released in 2013, and he received some success in country music formats due to his collaboration with Florida Georgia Line for their "Cruise" remix and his cover of Thomas Rhett's "Die a Happy Man." He has been a mainstay on BET for the last several years with the reality series Nellyville and the parody series Real Husbands of Hollywood.

Nelly's recent years have been mired with controversy. He was arrested in April 2015 after a traffic stop, when an officer smelled cannabis and searched his bus, finding handguns and a substance believed to be methamphetamine. The weapons and drugs allegedly belonged to a crew member on the bus. Nelly was booked and released on bail, charged with felony drug possession, simple possession of marijuana, and possession of drug paraphernalia. The felony charge was dropped after the substance was examined and confirmed not to be meth. Nelly pleaded guilty to the lesser charges and was sentenced to one year probation, with all convictions cleared after eleven months thanks to a diversion program.

In 2016, the IRS put a $2.4 million tax lien against Nelly for unpaid taxes.

In October 2017, Nelly was arrested in Auburn, WA, on suspicion of second-degree rape. He was released without charge, as the accuser declined to pursue the case, although she insisted he was guilty, citing a lack of resources to pursue the charges. Nelly was accused again of sexual assault in January 2018 by a UK woman. This investigation is pending. Nelly has denied the charges in both cases.

In addition to Nelly's businesses, he runs a nonprofit organization called 4Sho4Kids Foundation, as well as a music production school in St. Louis called Ex'treme Institute in cooperation with Vatterott College.
